# Nucleotides: why fast-dividing cells run short first

> The body builds nucleotides itself - but that synthesis is expensive, and gut lining and immune cells are worst at it. What «conditionally essential» actually means, and how WOO® Nukleotidmischung is set up.

Nucleotides are the building blocks of DNA and RNA - and therefore the precondition for a cell dividing at all. The body makes them itself. The catch: that endogenous synthesis is expensive, and the very cell types with the highest demand are the worst at it.

## Two routes to a finished nucleotide

**De novo synthesis** assembles nucleotides from scratch: from glutamine, glycine and aspartate, with ribose-5-phosphate as the sugar backbone. The pathway is long and costs considerable amounts of ATP.

The **salvage pathway**, by contrast, recycles bases already present from cell turnover or from food and puts them straight back to work - a fraction of the energy cost. The decisive point: enterocytes of the gut lining, lymphocytes and bone marrow cells have only limited de novo capacity. They depend on salvage - and therefore on a supply of nucleotides from outside.

## "Conditionally essential" - what that means

Under unremarkable conditions, endogenous production suffices. But when cell turnover rises - during infection, after injury, in growth phases, under heavy training loads or during caloric restriction - demand can outrun synthesis. Grimble and Westwood (2001) therefore classified nucleotides as **conditionally essential**: not always necessary, but limiting under stress.

The clearest signal comes from infant nutrition. Breast milk contains free nucleotides in relevant amounts; formula has been fortified accordingly since the 1990s. Studies on this showed effects on immune response and maturation of the gut lining (Carver & Walker 1995).

## The gut lining has the highest cell turnover in the body

Enterocytes are completely replaced every three to five days - no other tissue divides faster. That is precisely why the intestinal mucosa depends on a good nucleotide supply.

In animal models, a nucleotide-free diet led to flatter villi and delayed recovery after mucosal damage; adding nucleotides reversed it (Hess & Greenberg 2012). This cannot be transferred one-to-one to humans - but as a mechanism it is well described.

## Immune cells and hard training phases

Lymphocyte proliferation is a nucleotide-intensive process: every cell division demands a complete set of DNA. McNaughton et al. (2007) examined nucleotide supplementation under intense exercise and found higher salivary immunoglobulin A and a more muted cortisol response. These are small studies - indications, not proof.

## Frequently asked questions

**Don't I get enough from food already?**
Offal, fish, yeast and pulses are nucleotide-rich. A typical Western diet supplies rather little - estimates fall at one to two grams daily.

**Why these five in particular?**
They are the monophosphates of the four RNA bases plus inosine monophosphate, an intermediate in purine metabolism. That covers both base classes - purines and pyrimidines.

**How long should you take it?**
No acute effect is to be expected. Daily intake over weeks makes sense, especially during phases of increased physical load.
